The recent advancement of cryo-EM data analysis software packages substantially improved the results, producing higher-resolution maps and finding hidden structural elements or conformations. Among many cryo-EM data analysis software, RELION and cryoSPARC are the two most used packages because they are easy to use, the GUI is intuitive, and produce good results. In this report, we compared the two packages regarding their functionalities and suggested practical methods to transfer data between the two packages using PyEM script, csparc2star.py. The protocols we suggested here would help cryo-EM users to utilize the different and diverse functions of the two packages in a variety of ways.
